Abstract:
[objective] a microbial strain capable of transforming daidzein into S-equol was screened and identified. [Methods] under anaerobic conditions, the growth of non-target bacteria was inhibited by antibiotics and the strains were isolated by dilution plating method, the structure of the product was identified. Then, the phylogenetic tree of the bacterium was constructed by 16S rDNA sequence analysis, and the phylogenetic status of the bacterium was established by combining the morphology and colony characteristics of the bacteria. [Results] a gram-negative facultative anaerobe strain LH-52(JN861767) was isolated from rat intestine, which could convert daidzein into S-equol, the BLAST alignment of 16S rDNA sequencing results showed that the similarity between the strain and Proteus mirabilis reached 99%. Combined with the results of morphological characteristics and physiological and biochemical experiments, the strain was identified as Proteus mirabilis. According to the HPLC retention time, mass spectrometry, nuclear magnetic resonance spectroscopy data analysis, the product was identified as S-equol. [Conclusion] strain P.mirabilis LH-52 is the first screening of the conversion of daidzein to S-equol facultative anaerobic bacteria, compared with the literature reported strict anaerobic bacteria is more suitable for industrial production.

Abstract:
A UV analysis method for the determination of equol was established by using equol standard sample and human urine samples of edible Soybean isoflavone. The characteristics of the method and the reliability of the determination results were investigated. The results showed that the detection limit was 5 × 10-10g/mL and the quantitative limit was 15 × 10-10g/mL. The method has been applied to the determination of equol in human urine with a precision of 4.45% and a recovery of 100.25%. The results are reliable. The newly established UV method meets the requirements of quantitative analysis, and has the advantages of simple operation, fast and low cost, which is suitable for the determination of large quantities of samples.

Abstract:
Objective to observe the effect of equol on osteoporosis in ovariectomized rats. Methods female SD rats were ovariectomized to establish osteoporosis model. Healthy 3-month-old female SD rats were randomly divided into five groups: sham Operation Group (sham), ovariectomized group (OVX),OVX + equol high dose group (equol, eq 0.5 mg/kg.d),OVX + equol low dose group (equol,Eq 0.25 mg/kg). d),OVX + estradiol positive control group (estradiol,E2). After 8 weeks of ovariectomy, the right femur was taken for bone mineral density (BMD) after 8 weeks of treatment, and blood was taken from eyes for blood calcium, serum phosphorus and osteocalcin (bone gla protein,BGP) content; ELISA kit for detection of serum estrogen (ES), osteoblast osteoprotegerin (OPG) and osteoclast differentiation factor (receptor activator of nuclear factor kappa B ligand,RANKL) levels; q RT-PCR method to detect the expression of OPG and RANKL mR NA. Results compared with the sham group, the ES value, BMD and blood calcium level in the OVX group were significantly decreased (P0.05), which were 78.1%,84.4% and 83.2% of the sham group. At the same time, the levels of serum phosphorus and BGP in OVX group were significantly increased (P0.05), indicating that the osteoporosis model was successfully established after ovariectomy, the expression of OPG protein and OPG mR NA decreased significantly, and the expression of RANKL protein and RANKL mR NA increased significantly (P0.05), after treatment with Eq and E2, the expression of OPG protein and OPG mR NA was significantly increased, the expression of RANKL protein and RANKL mR NA was down-regulated, and the BMD and blood calcium level of OVX rats were increased, decreased blood phosphorus and BGP content (P0.05). Conclusion equol can effectively improve osteoporosis in ovariectomized rats, which may be related to the up-regulation of OPG and inhibition of RANKL secretion.expand

Abstract:
equol is an important metabolite of soy isoflavones, about 30% to 50% of the population can convert soy isoflavones into equol and then excretion, however, the factors affecting its transformation are not completely clear, and intestinal flora may be the most important factor. Equol has more effective biological effects than its prototype Soybean isoflavone. Research and development of the biological activity of equol, the law of metabolic transformation and its physiological function, for the prevention and control of cardiovascular disease, osteoporosis, dyslipidemia, obesity, menopausal syndrome and breast cancer, prostate cancer and many other chronic diseases are of great significance. In this paper, the metabolic process of equol in human body, biological activity and preventive effect on various chronic diseases are reviewed.

Abstract:
The aglycones of soybean isoflavones are mainly genistein and coumarin, which can prevent and improve various diseases. In recent years, a large number of studies have shown that the biological effects of soybean isoflavones are partly attributed to equol. Daidzein is converted to equol by specific flora in the gut, and not everyone can produce equol. In this paper, the bacteria related to the production of equol in human and animal intestines are reviewed, and the strains that can directly convert soybean to equol are introduced. Equol-producing related bacteria can produce equol by anaerobic culture in vitro.
